Inhibition by acarbose is demonstrated for human α-amylase using maltoheptaose (Mal-7), and sucrase, maltase and isomaltase activities, using sucrose, maltose and isomaltose, respectively. Sucrase-isomaltase was extracted from cultured human enterocytes for assessment of α-glucosidase activities. Data in sheet 6E show acarbose IC50 values may be affected by substrate concentrations, therefore assay optimisation is necessary. Data represent mean ± SD from a batch of samples of n = 3, with duplicate injections per sample, with R2 ≥ 0.99.
