MyNon24Sleep v2

Published on by Stephen Karl Larroque

MyNon24Sleep v2 is a pilot case study for the design of a Remote Decentralized Clinical Trial (RDCTs) for the 24/7 study of human circadian rhythm disorders over long periods of time using off-the-shelf non-invasive medical grade accuracy wearables.

MyNon24Sleep v2 is the continuation into the 2nd year of the MyNon24Sleep v1 project, with additional sensors and modified positioning and fixation systems for others, to improve robustness of data collection under any free living conditions. This means that this v2 dataset has much fewer gaps than v1, and the data collection was done under all kinds of environments, including regular outdoors activities, whereas v1 was constrained nearly all the time at home to better control factors. The v2 is hence an extension of the v1 experiment to a broader set of conditions and environments with different bright light pattern exposures and physical activities, on the same subject.

You can find the v1 dataset here: https://figshare.com/projects/MyNon24Sleep_-_A_self-study_of_the_circadian_rhythm_and_its_altering_factors/101804

The data collection for v2 ran from March 2022 to March 2023. Note that beyond this date, the data collection continued, but because of several sensors breaking and no funding but my own, the experiment has since been halted (and it may be resumed later but at an unknown date).

Since some of the sensors did not change (eg, ECG, core body temperature, wrist skin temperature), these data can be concatenated between both v1 and v2 datasets to make a 2 years dataset, which can be an interesting opportunity for longitudinal analyses.

This v2 dataset includes the following biomarkers:

* Manually curated sleep diary to track sleep-wake patterns and provide additional context with tags and free-form comments over controlled factors.

* Core body temperature measuments (non-invasive) tracked continuously 24/7 at 1Hz using the GreenTEG CORE device. Placed axially, with temperature sensing ~2cm below skin, hence the core body temperature estimated here should be considered an approximation of pulmonary temperature, not rectal (same for v1).

* ECG and 3-axis trunk actigraphy tracked continuously 24/7 at 130Hz and 100Hz respectively using Polar H10.

* 6-axis actigraphy tracked continuously 24/7 at 100Hz using Axivity AX6.

* Wrist skin temperature on the radial artery in the forearm tracked continuously 24/7 with a sampling rate of one sample every 5min using Maxim Thermocron iButtons. A dorsal iButton (on the other side of the forearm) is also worn to serve as a functional control since it does not capture the radial artery.

* Lys Button, a circadian light sensor (ie, intensity and color, with internal calculations of melanopic lux mLux).

The v1 dataset includes other datasets that are still applicable but not repeated here, such as MRI.

More information on the experiment's design, the wearable devices used for data acquisition and some data analysis tools can be found under an open-source licence in the Wearadian GitHub repository, which was updated to include a description of the changes for v2: https://github.com/Circadiaware/wearadian

 Additional infos about the subject:

* Height: 1m74.

* Weight: Between 60 and 65 kg during the whole experiment period (regularly updated weight measures have been added as a dataset).
