(A) The addition of unanchored K63-linked polyUb chains enhances WT VP35 interactions with NP. Lysates from HEK293T cells expressing WT-VP35 were mixed with lysate from cells expressing NP, in the presence or absence of added purified unanchored K63-linked polyUb chains [212], followed by coIP with anti-Flag beads. (B) Treatment with pCEBS or SFC in a minigenome assay reduces interactions between VP35 and NP in the co-immunoprecipitation assay. HEK293T cells were transfected with minigenome components and 4 h post-transfection cells were treated with 200 μm of pCEBS and SFC; 50 h later cells were lysed, and immunoprecipitation assay was performed using FLAG beads.
