Immunofluorescence
labeling results (Supplementary Figure 1)
show that isolated vaginal cells express Myosin Heavy Chain 11 (mature SMC marker),
Fibroblast-Specific Protein-1 (FSP-1), but not pan-cytokeratin antibodies
(AE-1/ AE-3) that are expressed by epithelial cells. These results together with earlier published findings that the cells express alpha smooth muscle actin, caldesmon and tropomyosin, classic smooth muscle cell markers, conclusively
show that the vaginal cells are composed of SMCs and fibroblasts for which
reason they are termed as non-epithelial vaginal cells (NEVCs).
