WT C57BL/6 and IL17RA-/- (KO) mice were infected with 10 μl of PR8 (50xTCID50) or PBS on day 0. Mice were euthanized at 7 dpi. for cell phenotyping and cytokine analysis. A-B. Neutrophils (CD11b+Ly6C+Ly6G+) present in nasal lavage and expressed as % of CD11b+ cells. C. CXCL1 chemokine levels in nasal lavage. D. Macrophages (CD11b+F4/80+) and monocytes (CD11b+Ly6C+Ly6G-) counts in nasal lavage. E. Neutrophils (CD11b+Ly6G+) counts in blood. F. Cytokine levels in nasal lavage determined by flow cytometric bead array. Data are representative of two independent experiments (n = 3-4/experiment) and expressed as SEM. For lavage cell phenotyping and cytokine analysis, samples were pooled and split into triplicate. Data were analyzed by One-Way ANOVA with Tukey’s post-hoc. *p<0.05, **p<0.01, ***p<0.001; ns, not significant.
