Generation and characterization of cancer-associated fibroblast cultures.

Fibroblasts were extracted from the resected tissues of two pancreatic ductal adenocarcinomas. The purity of the cultured fibroblast cells was verified through immunofluorescence to detect Pan-CK (red), E-cadherin (green), vimentin (red), α-SMA (red), PDGFR-β (red), CD31 (red) and D2-40 (red). The cell nuclei were counterstained with DAPI (blue). Positive control tissues for CD31 and D2-40 immunostaining have been used (Figure S1). B. Western blot analysis confirmed the expression pattern of the markers used in A.
