(A) Bone marrow cells from wild-type (Fyn+/+) or Fyn knockout (Fyn−/−) mice were transduced with vector (GFP) or vector expressing p210 BCR-ABL1 (p210). Expression of p210 and Fyn was detected in total protein lysates (100 µg) using antibodies specific for Abl and Fyn, respectively. Actin was used as a loading control. The blot is representative of three independent experiments. (B) Transduced bone marrow cells were plated in methylcellulose soft agar and colonies were counted after five days. *p<0.01, n = 3, mean+SEM. (C) Transduced bone marrow cells were plated in 96-well plates at 150,000 cells/mL. Every two days, cells were stained with trypan blue and viable cells were counted to determine growth rate. *p<0.001, n = 5, mean+SEM.
