Then the each diluted sample was partly counted by Fluorescence microscope with SYBGreen as fluorescence stain (X axis). Another part were determined by Spectrophotometer under the Ex (Excitation wavelength) = 440 nm and Em (Emission wavelength) = 680 nm, the determined value, Fluorescence intensity (Y axis) could indirectly reflect the numbers of the cells.
