These files contain a physiological analysis of streptomycin and tailocin treated Pph1448A. We used a combination of metabolic reporter dyes (RSG, PI, and Hoechst 33342, or RSG, Draq7, and Hoescht). Pph cells were stained with these dyes and images were taken under fluorescence-phase contrast microscope.

Experiments were conducted at log and stationary phase, with tailocin or streptomycin, and before and at one timepoint after treatment. Streptomycin is abbreviated as Strep and Tailocin as Tail in filenames. 10 fields were randomly chosen and imaged for each growth phase, treatment, and timepoint (80 images per experiment). Each experiment was conducted four times on four separate days (320 images total).

In an experiment to evaluate DRAQ7 staining depicted in Supplemental Table 2, we imaged stationary phase only with treatments of streptomycin and tailocin at two timepoints (4 treatments X 10 fields = 40 images total)

Due to the large size of the .czi multichannel image files(>50gb) we are only uploading the .jpgs of the merged images, but we are happy to provide any multichannel images if there is interest.

Images used to categorize cells into categories 1 to 5 for Figure 2 and supplementary figure 5 are labeled with the convention Growthphase+TreatmentTrial#Timepoint#(Image#) as follows:

Stationary phase Pph:

Stationary+StrepTrial1T0(1-10),

Stationary+StrepTrial1T5(1 to 10),

Stationary+TailTrial1 to 4T0(1 to 10),

Stationary+TailTrial1 to 4T4(1 to 10)

Log phase Pph:

Log+StrepTrial(1 to 4)T0 (1 to 10)

Log+StrepTrial(1 to 4)T5 (1 to 10)

Log+TailTrial(1 to 4)T0 (1 to 10)

Log+TailTrial(1 to 4)T4 (1 to 10)

Merged images of multichannel files used to calculate for supplementary table 2, which depicts RSG, DRAQ7 and Hoechst33342 stained stationary phase Pph, are labeled as Stationary+StrepDRAQ7 T0(1 to 10), Stationary+StrepDRAQ7 T5(1 to 10) Stationary+TailDRAQ7T0(1 to 10), Stationary+TailDRAQ7T4(1 to 10)
