WitmerLab at Ohio University has been experimenting with methods to increase soft-tissue contrast in microCT scans of relatively large vertebrate specimens. We have used diceCT very successfully for years—and continue to do so—but it has some shortcomings owing to the often long diffusion times required for large-ish specimens. SpiceCT involves perfusing vertebrate specimens (i.e., injecting through the blood vascular system) with Lugol's iodine (I2KI). We typically inject fresh or thawed (i.e., unfixed) specimens because the tissues are more compliant than when fixed. Staining is basically instantaneous. As a result, specimens can go from start to finish—raw carcass to analyzing CT data—in a matter of hours.
