A. THP1-Xblue cells were transfected with a siRNA targeting XIAP (siXIAP) or with a non-target siRNA (siCTRL). 48 h post transfection, proteins were extracted. Samples were immunoblotted with an anti-XIAP antibody. Actin was used as a loading control. B. THP1-Xblue cells were transfected with a siRNA targeting XIAP (siXIAP) or with a non-target siRNA (siCTRL). 36 h post infection, cells were treated for 16 h with MDP (10 or 25 µg/mL) or with TNF-α (500 U/mL) or left untreated. NF-κB activity was measured by colorimetric enzyme assay (mean ± S.D (n = 3), *p<0.004). C, D, E. THP1-Xblue cells were transfected with a siRNA targeting XIAP (siXIAP) or with a scramble siRNA (siCTRL). 48 h post transfection, cells were treated with MDP (100 µg/mL) or with TNF-α (500 U/mL) for indicated periods. RNAs were extracted and il-8, il-6 and tnf-α mRNA levels were measured by quantitative real-time PCR (Mean ± S.D (n = 3), *p<0.0001).
