(A) Representative methylation patterns of medulloblastoma samples, with samples organized by row and each CpG location organized in columns. Leftmost column indicates OTX2 expression status (red, OTX2-expressing; green, OTX2-nonexpressing), letters to the right of this column indicate OTX2 copy number status (N, normal; G, gained; HL, heterozygous loss), if known [5], [19], [32], [33]. (B) OTX2 mRNA level, as determined by RT-qPCR, in tumors exhibiting either a methylated or unmethylated OTX2 promoter. ND, not detected. (C) Semi-quantitative RT-PCR of OTX2-nonexpressing cells treated with the indicated concentrations of 5′-aza-2′-deoxycytidine for 5 days. D283 medulloblastoma cells serve as a positive control for OTX2 detection.
