The assay was performed with 260 nM PyAeADHII in 50 mM NaPO4 pH 7.5, 3 mM α-tetralone, 0.3 mM NADPH in the presence of 80–1200 µM flunarizine dihydrochloride in DMSO/isopropanol. a) Absorbance of NADPH (340nm) was measured over time at different flunarizine concentrations. b) The rate of NADPH conversion (Δa.u. = absorbance unit change/minute) versus flunarizine concentration.
