Shown is an immunoblot obtained under optimal conditions for the antibody against myosin light chain kinase (MLCK). Each immunoblot included lanes loaded with varying known amounts of a pooled standard, which enabled construction of standard curves that allowed calculation of unknown masses via non-linear regression. All lanes with unknown samples represent equal amounts of total protein loaded from homogenates of normoxic (FN) and chronically hypoxic (FH) fetal lamb arteries under multiple experimental conditions. Fresh homogenates indicate preparations made the day of harvest, and the adjacent bands represent homogenates organ cultured for 72 hours under either normoxic (21% O2) or hypoxic (1% O2) conditions. The molecular weight markers confirmed that the protein detected exhibited a single band at a mass of ≈150 kDa, as expected.
