<p dir="ltr">A panel of 7 chordoma cell lines was plated in a white-walled 96-well plate to evaluate response to ART0380, an ATR inhibitor, as a single agent treatment and to evaluate synergy in combination with SN38. treatment using CellTiter-Glo. All cell lines were plated so that at treatment start, 36 hours after plating, the confluence was 15%, and the assay endpoint for each cell line was time to 80-90% confluence for untreated cells. Experiments were repeated for each cell line at least once to ensure reproducibility. Representative curves and absolute IC50s are shown. Cell line-specific seeding densities and assay durations can be found through the link at the bottom of this page. All lines were plated in 100uL of (4:1) IMDM/RPMI 1640 + 10% FBS + 1% NEAA + 1% GlutaMAX. The top ART0380 concentration used in the assay was 10uM, diluted by 1/3rd to the lowest concentration of 1.52 nM. The top SN38 concentration used in the assay was 1uM, diluted by 1/3rd to the lowest concentration of 0.152 nM. In the combination treatment, the top ART0380 concentration was 10 uM, diluted by 1/3rd to the lowest concentration of 1.52 nM while the concentration of SN38 was held constant at 3 nM for CH22 and U-CHCF365 and 30nM for UM-Chor1, UM-Chor5C, U-CH1, U-CH17PII, and U-CH2. SN38 concentrations used in the combination treatments were determined based on each cell line's sensitivity to SN38, which can be found in each graph as the grey cure indicates single agent SN38. The viability at each concentration was measured in triplicate for each dose-response curve. Plotted are the mean values +/- the standard deviation. The data for single agent treatments was normalized to the DMSO-only control.
