Hela cells were treated in triplicate with anti-miR-21 ASOs at 33 nM containing a number of 2′-sugar and backbone modifications. At 24 h post transfection, cells were lysed and anti-miR-21 activity measured using the luciferase reporter assay. The 2′-sugar and backbone modification of each anti-miR-21 ASO is indicated to the left of the graph, and each compound is referred to by the indicated number in the text. O, 2′-O-methyl; M, 2′-O-methoxyethyl; D, 2′deoxy; underline denotes phosphorothioate modified backbone.
