(A) Ovarian cells were transfected for 24 hours with plasmid encoding HSP90B1; cells transfected with empty vector served as the control. At 48 hours post-transfection, the expression of HSP90B1 was analyzed by Western blot analysis, and GAPDH antibody was used as a loading control. One representative result is shown. (B) The cells transfected with plasmid encoding HSP90B1 were used for the cell viability assay using MTT. Data are presented as the mean cell viability ± standard error, n = 10. (C) The transfected cells were analyzed for apoptosis by flow cytometry after staining with propidium iodide (PI) and Annexin V. Data are presented as dot plots, and one representative result is shown. (D) Apoptotic cells were analyzed quantitatively, and the data are presented as the mean percentage of apoptotic cells ± standard error, n = 10. (*P<0.05, compared with Ctrl-vector; #P<0.05, compared with PCOS-Vector).
