The purpose of the experiment was to isolate fluorescently tagged cells from single cell suspensions of Drosophilamelanogasterembryos aged to stage 10-12. Cells were expressing a fluorescently tagged protein localising to the plasma membrane in either the salivary gland primordium only (SrcGFP) or in all of the embryonic epithelium (ArmYFP). The isolated fluorescent cells were sent for single cell RNA sequencing to investigate the genetic programmes involved in early salivary gland tubulogenesis.
