Raw dose-response growth curves to cisplatin of various breast cancer cell line models. Corresponding results files calculating drug sensitivity parameters according to Hafner et al. 2016.

File names in ‘Parameters’ files

‘[…]_CellNr’ / ‘[…]_Conf’ Analysis performed on Fluorescent Channel (Cell Number) or Confluence Channel (Conf).‘[…]_1/2/3’ Analyzed replicate --> replicate whose initial confluence is closest to 20%.

Excel sheets in ‘Parameters’ files

Fitting of 4-day dose-dependent growth curves to exponential function:

  • ‘exp_k’ exponential growth rate for each condition (fitting of 4-day growth curves to exponential function)
  • ‘exp_CIlow’ / ‘exp_CIup’ confidence intervals of the exponential growth rate.
  • ‘exp_Rsq’ Rsq-value of each exponential fit.

Calculation of doubling times from final and initial values of the time-series:

  • ‘doublingtime’ doubling time in hours
  • ‘doublingtime_std’ standard deviation of doubling times, propagated from standard deviation across two technical replicates.

Fitting of sigmoidal function to dose-dependent exponential growth rates (as described in Hafner et al.,2016)

  • ‘parameters’ cleaned growth rate inhibition parameters —> “drug-sensitivity parameters”
  • ‘parameters’ dose eliciting a GRinf response (not used in the manuscript but extracted alongside).
