RNA sequencing of cultured HMC3 cell lines in vitro. Human Microglia Clone 3 cell lines were treated with oxygen and glucose deprivation (OGD) for 4h and reoxygenation for 24h, which was performed as an in vitro model of ischemic stroke as previously described. At the beginning of reoxygenation, HMC3 cell lines were separately treated with exosomes isolated from LAA patients or exosomes isolated from healthy individuals at the concentration of 20 μg/mL. After reoxygenation, total RNA from HMC3 cell lines was extracted for cDNA synthesis, library construction and sequencing. Procedures for data analysis were similar with that of sorted microglia in vivo.
