This is a method figure showing how the Clontech smRNA kit works to polyadenylate miRNAs, uses a modified Moloney Murine Leukemia Virus for first strand synthesis, and the first-strand leaves a non-templated oligo C (three cytosines). The second strand occurs by template switching using the 3C for priming. The resulting fragments can be indexed by PCR using standard TruSeq primers.
