Splenic mDCs are the major IFN-αβ-, but not the predominant IL-6-producing cells in adenovirus infected mice.

<p>Ad-induced IFN-αβ and IL-6 responses in mice depleted for mDCs and pDCs. Groups of wt and CD11c-DTR mice (4/group) were injected with DT i. p. or left untreated. 24 h later the animals were injected with 1.2×1010 Ad3 particles and the plasma levels of IFN-αβ (A) and IL-6 (B) were measured 4 and 8 h after infection. Mice (4/group) were untreated or injected with 500 mg of anti-mPDCA-1 antibody i. p. and challenged with 1.2×1010 Ad3 particles. Plasma IFN-αβ levels were determined 8 h after Ad infection (C). The efficacy of splenic DC depletions is shown in <a href="http://www.plospathogens.org/article/info:doi/10.1371/journal.ppat.1000208#ppat.1000208.s005" target="_blank">Fig. S5</a> and <a href="http://www.plospathogens.org/article/info:doi/10.1371/journal.ppat.1000208#ppat.1000208.s006" target="_blank">S6</a>.</p>
