Prox1 represses CYP7A1 transcription and bile acid synthesis in HepG2 cells.

(A) Prox1 represses transcription of CYP7A1 mRNA. HepG2 cells were co-infected with recombinant lentiviruses expressing Prox1-targeting siRNA precursors si258 or si1646, or scrambled control siSCR, and recombinant lentiviruses expressing control GFP or siRNA-insensitive Prox1 mutant Prox1m as indicated. Total RNA was extracted 36 hrs post-infection and levels of CYP7A1 mRNA measured using quantitative real-time PCR as described in Materials and Methods. Means and SD from three independent experiments are presented. Prox1 expression levels were analyzed in Western blot using beta-actin as loading control (top). (B) Prox1 represses BA synthesis. HepG2 cells were infected with recombinant lentiviruses expressing Prox1-targeting siRNA precursors si258 or si1646, or scrambled control siSCR. Total intracellular BA and phospholipids were extracted and measured as described in Materials and Methods. Relative bile acid levels are expressed as BA/phospholipids and presented, taking result from lenti-siSCR-infected cells as 1. Means and SD from three independent experiments are presented. Statistically significant changes (P<0.05 in student’s t test) were indicated (*).
