Generation of Sox2-CreER BAC transgenic mice.

(A) Illustration of the transgene construction. In the BAC clone RP23-2B8, the Sox2 coding sequence was replaced by CreER™ sequence. The two black boxes represent the 5′UTR and 3″ UTR of the Sox2 gene. H, HindIII; B, BglI. (B) Positive founder lines (No.20, 34, and 36) were identified by PCR and Southern blot. For PCR, Cre sequence was amplified. For Southern blot, genomic DNA was digested with HindIII and BglI and probed with a Cre sequence.
