(A) Levels of cellular cAMP following SeV infection. HeLa cells were infected with SeV (MOI = 1) for the indicated times. Cell lysates were analyzed by cAMP enzyme immunoassay kit. (B) Levels of cellular cAMP following SeV infection in Rig-I+/+ and Rig-I-/- MEFs. The indicated MEFs were infected with SeV (MOI = 1) for the indicated times. Cell lysates were analyzed by cAMP enzyme immunoassay kit. (C) SeV infection induces PKACs phosphorylation. HeLa cells were infected with SeV (MOI = 1) for the indicated times before immunoblot analysis with the indicated antibodies. (D) Effects of exogenous cAMP on SeV-induced transcription of IFNB1 gene. HeLa cells were transfected with PKACs RNAi plasmid and selected with puromycin, then infected with SeV (MOI = 1) and treated with cAMP (1x107 pmol/mL) or left untreated for 6 h before qPCR was performed. The right blots show the expression levels of PKACα/β.
