(A) Murine 10.1 fibroblasts were infected with the 3ˈ M117 or m117.1 HA-tagged viruses at an MOI of 1 TCID50/cell. Cell lysates were harvested at the indicated times post infection and subjected to SDS-PAGE. 20 μg of protein were loaded. MCMV E1 and cellular β-actin served as infection and loading controls, respectively. (B) Murine 10.1 fibroblasts were infected with the WT MCMV or the 5' M117 HA-tagged viruses at an MOI of 1 TCID50/cell and cell lysates were harvested 24 hpi and subjected to SDS-PAGE. (C) Cells infected with MCMV-M117-HA were treated for 4 hours with 50 μg/mL cycloheximide (CHX) followed by 5 μg/mL Actinomycin D (ActD) or no (–) treatment for 7.5 hours prior to western blot analysis. (D) Cells infected with MCMV-M117-HA were treated for 24 hours with 250 ng/mL phosphonoacetic acid (PAA) prior to Western blot analysis.
