Live cell imaging of PC3 prostate cancer cells after GFP-EN2 transfection and the production of EN2-containing microvesicles

Using a confocal microscope and glass-bottom dishes, PC3 prostate cancer cells were transiently transfected with GFP-EN2 and imaged every 5 minutes up to 24 hours. The confocal microscope was chambered, which allowed the cells to be incubated at 37 degrees and supplemented with CO2.

The GFP-EN2 is secreted in large vesicles, under a seemingly regulated process. The vesicles are made intracellularly and not at the membrane.

This was carried out at The University of Surrey, with support from the Ringrose Family Trust, as part of my doctoral degree 'Investigating engrailed-2 (EN2) as a therapeutic target in prostate cancer'. The full theses and associated open-access publication are linked below.
